Abstract : Channel catfish and common carp embryos were microinjected with mouse metallothionein human growth hormone fusion gene (MThGH), two constructs of RSV rainbow trout growth hormone cDNA (pRSVrtGHcDNA), RSV coho salmon growth hormone cDNA (pRSVcsGHcDNA) and rainbow trout vitellogenin gene (rtVG20) at the early, intermediate and late one-cell stage and at the two-cell and four-cell stages to determine the developmental stage appropriate for maximizing production of transgenic fish. Survival (hatching rate) of microinjected channel catfish embryos relative to non-injected controls was 65.0, 43.6, 38.5, 27.4 and 35.9% for early, intermediate and late one-cell, two-cell and four-cell stages, respectively. Survival of microinjected common carp embryos relative to non-injected controls was 88.9, 85.3, 91.5, 85.3 and 83.4% for early, intermediate and late one-cell, two-cell and four-cell stages, respectively. Twenty-two of 420 (5.2%) microinjected channel catfish analyzed contained sequences of foreign DNA, and the frequency of integration was 2.1, 1.5, 15.2, 12.5 and 7.7% at the early, intermediate and late one-cell, two-cell and four-cell stages. Twenty-five of 533 (4.7%) microinjected common carp contained sequences of foreign DNA and the frequency of integration was 8.3, 1.0, 0.0, 11.5 and 1.7% at the five respective developmental stages. All transgenic common carp (pRSVrtGHcDNA-construct 1) produced trout GH polypeptide. The quantity of growth hormone detected ranged from 8.0 ng/mg to 89.1 ng/mg of protein in red blood cells. Ninety-day-old transgenic common carp that developed from microinjection of pRSVrtGHcDNA at early one-cell stage grew 20% faster (P < 0.05) than normal siblings. Those transgenic individuals microinjected at the two-cell stage grew at the same rate as normal siblings. Three of 4 male transgenic common carp tested transmitted the foreign DNA to their progeny. Positive F progeny grew 20-40% faster (P < 0.001) than their normal siblings, and 32-46% of these individuals were larger than the largest control at 90 days of age.
