Abstract : The progressive changes in the chromosome alignment processes in wild type and ahp2 mutant of Arabidopsis thaliana (Ler ) have been studied using the serial ultrathin-sectioning technique for transmission electron microscopy. This technique preserves weak alignments between chromosome axes as well as synaptonemal complexes (SC), and has allowed identification of at least two other structures previously not described in Arabidopsis meiocytes. The ahp2 mutant has severely reduced SC, resulting in only 5.1% SC, compared to 84% in wild type. It is clear that the ahp2 mutant can undergo rough alignment (44.0% of total bivalent length) and does convert appreciable amounts of the rough alignment into close alignments (39.0% of the total bivalent length). Thus, we have proposed that the Ahp2 mediated strand invasion may be required once rough alignment occurs and may facilitate close alignment. Lack of Ahp2 may delay close alignment and prevent appreciable amounts of SC.
