ABSTRACT<br>Shifaa Alshammari. Cytotoxic Activity of Ferula assa-foetida and Astragalus sarcocolla Against Cervical Epithelial and Colon Carcinoma Cell Lines (under the direction of Dr. E. Lewis Myles).<br>Cancer is amongst the leading causes of death across the globe. Conventional ways of cancer therapy are extremely harmful to human health. Therefore, many investigations focus primarily on developing remedies from herbs as a source of cancer treatments. In this study plants from Iran, which include Astragalus sarcocolla and Ferula assafoetide have been tested for cytotoxicity activity on HeLa and SW620 cell lines. I hypothesized that Astragalus sarcocolla and Ferula assafoetide would show the best anticancer growth. This study involves methanolic extracts of Astragalus sarcocolla and Ferula assafoetide and aqueous extract from Astragalus sarcocolla. Methanol and aqueous extracts were tested against human metastatic cell lines representing cervical cancer (HeLa) and colorectal cancer (SW620) cells for 24 hours. Growth analysis was then determined using a cell viability indicator Alamar blue and a florescent plate reader. As a result, the study found that the extracts from the Ferula assafoetide has shown anticancer growth against HeLa and SW620 the cell lines at the highest concentrations (12,11, 10 and 9μg /μl). Diluted concentrations have demonstrated good inhibitory effects with IC50 values ranging between 95-55% inhibition. Analysis of the methanolic extracts of Astragalus sarcocolla on HeLa and SW620 cell lines show results of anticancer growth.<br>The inhibition was observed at concentrations (0.455 μg /μl). The IC50 values ranges from 85-95% inhibition. On the other hand, analysis of the aqueous extract of Astragalus sarcocolla has shown anticancer growth against HeLa and SW620 cell lines at all concentrations. Concluding, that methanolic extracts from Astragalus sarcocolla and Ferula assafoetide and aqueous extract of Astragalus sarcocolla should be studied further to isolate the active antitumor compounds in the extracts and to investigate the mechanism of action of the compounds.
